Aegean and Olympic airways have multiple flights daily from Athens to Mykonos. There are also flights from Thessaloniki. There are weekly charter flights from Italy, the UK, France, and Germany and less frequent charters from other Western European and international airports.

Ferries:

Daily ferries from Pireaus Port of different varieties, high speed 3hrs and standard ferry 6hrs.

Car Rentals:

Avis, Hertz, Europe Car and smaller agencies that I can recommend if there is interest.
